Foci detection

Hello,

I am trying to analyze the localization of a fluorescently tagged protein inside bacterial cells using MicrobeJ. However, the Maxima detection is not working at all. What puzzles me is that I was able to analyze the exact same type of images with this method a few years ago without any issues, but now it no longer works. Would it be possible for me to share an image file so that you could help identify the cause?
