Some questions....

 

1) When we are saying that particular allele is recessive, But because of what it is recessive? Is it because of mutation in regulatory sequences of that allele or because of mutation in that allele sequence?

 CERTANLY ONLY ONE ALLELE OF ALTERNATE FORMS EXISTING ONE ON EACH CHROMOSOME COMPLEMENT EXPRESS.  THE ALLELE THAT DOES NOT MAKE IT UP FOR FUNCATIONALITY IS RECESSIVE.  THE LEVEL AT WHICH THE THE RECESSIVE ALLELE REGULATED IS BELIVED TO BE VARYING-IT COULD BE TRANCRIPTION (LARGELY), TRANSLATION, POST TRANSCRIPTION, AND EVEN POST TRANSLATIONAL MODIFICATION TO THE PROTEIN-ALL ARE IMPLICATED.  DEPENDING ON WHICH OF THESE PHENOMENA IS OPERATING, THE MUTATION SITE OR THE FACTOR RESPONSIBLE FOR REGULATION WOULD EXISIT, IT COULD BE RIGHT IN THE ALELLE CODING SEQUENCE, IT COULD BE IN PROMOTER SEQUENCE, OR REGULATOR SEQUENCES, ENHANCER SEQUENCES, REGULATING SMALL RNAs ASSOCIATED THE GENE ETC., 

I WAS PART OF ONE PROJECT DBT MILLET BIOTECHNOLOGY NETWORK WHERE IN BAJRA, DIFFERENE IN PROMOTERS OF GENES OF SINGLE FAMILY WAS SHOWN TO BE RESPONSIBLE FOR DIFFERETIAL EXPRESSION-ESSENTIALLY SNiPs…HOPE I ANSWERED YOUR QUESTION

 

2) If we clone the mutant allele with constitutive promoter and do the expression in transgenic, what will be status of that partucular allele? It will behave like dominant or recessive?

 

DEPENDS ON WHERE THE ROUTECASUE FOR RECESSIVITY EXISTS-IF NOT IN CODING REGION, I DO NOT SEE ANY REASON WHY IT SHOULD NOT BE DOMINANT/EXPRESS IN TRANSGENIC WITH CORRESPONDING PHENOTYPE
