Just asking as I am having issues with certain parameters being unavailable.

Info Service 15:41:29.594-04:00 on 2018-10-18 Calling function of DirectMethodAccess: SetCustomScan(0.5;1003 FirstMass=50.00 LastMass=852.35 IsolationRangeLow=401.48 IsolationRangeHigh=402.88 MsxInjectRanges=[] MsxInjectTargets=[] MsxInjectMaxITs=[] MsxInjectNCEs=[] MsxInjectDirectCEs=[] MaxIT=28.00 Resolution=17500 Polarity=0 NCE=27.0 NCE_NormCharge=2 NCE_SteppedEnergy=0 NCE_Factors=[] HCDTrappingTime=3.00 DirectCE=0.00 SourceCID=0.00 Microscans=1 AGC_Target=100000 AGC_Mode=1 CTCD_Support=1 SLensRFLevel=40.00)

I realize this is the "boxcar" method, but wasn't expecting so many boxes in the log file.  e.g., MsxInjectRanges=[] MsxInjectTargets=[] MsxInjectMaxITs=[] MsxInjectNCEs=[] MsxInjectDirectCEs=[]

QE classic running Tune2.9SP1 32bit

Thanks,
JP

Dear JP,

MQ.Live has not been tested on a QE classic.

Could you please provide further detail which parameters are unavailable? The status log appears ok.

it works beautifully here on all our QE classics, Tune 2.9. No extras needed, just change the res settings etc. to match QE classic standards.

best,
Erwin

Hey Erwin,

I have tried it on Q Ex instrument as well and it works smoothly. However, I am not getting any improvement in comparison to the Top12 method that I am using on the same instrument (I am using the Kelstrup fast/sensitive methods). Do you get any improvement in term of peptides and protein groups IDs?

Hi Yaarub,

We're also not really getting improvements in terms of protein IDs compared to standard DDA on a single-shot complex lysate analysis, but I think the power of BoxCar will really show itself when combined with some sort of spectral library. We are testing at the moment and am looking forward to seeing how it pans out. Also just goes to show how good the QE and Kelstrup methods are for DDA, that such a classic instrument is still able to represent itself amongst the newer generations! :)

cheers,
Erwin

Hey Erwin,

Thanks for your reply.
i am also preparing a peptide library to test it with BoxCar. I have finished acquiring all the runs, I just need to find time to analyze the data in the next 2 weeks. I will keep you updated. it would great if you could also post something here in case you have an update.

Hi Erwin,

Our lab is also trying to get MQ-Live working on our QE classic. May I ask what .Net version you are running with the machine? We are on 4.6.2 which doesn't seem to play well with the newest version of MQ-Live, but we wanted to check in to see if .Net 4.7 plays nicely with the QE classic before upgrading.

Thanks,
Anthony
