Basically my map window is displaying exactly as I want it, as is my
key.

However when I go to the layout window, two layers containing building
points don't display at all there is a gap in the key where these
layer titles should be.

Has anyone experienced this?

It's a workspace which I haven't used for a couple of months and was
really hoping to avoid having to redo all the label positioning. I
can only think it must be a bug?

First, make sure the frame is linked to the correct Map window. Might the
workspace contain another map window that is almost like the one you want to
show, but missing the point layers?

Once you've done that, check to see if the building point layers are zoom
layered or not. You shouldn't use zoom layering in a map you intend to put
in a layout window. Such Map windows should contain only the layers that
you want to see in the layout.

MapInfo's zoom layering is tied to particular zoom values (i.e. east-west
distance across the window or frame) rather than to scale values. Zoom
isn't really the right criterion to use, but what can you do?

Because of this, the map's zoom in the layout frame isn't necessarily the
same as it is in the window, and zoom-layered layers that appear in the Map
window might not appear in the layout, and vice versa.

Finally, if you're using labels rather than text objects, you will almost
certainly end up repositioning every one of them if you change the map
scale.
