What is Cenforce?
Cenforce
consist of sildenafil citrate. It is prescribed for the treatment of
persistent erectile dysfunction (ED) in men. ED can be defined as
inability to achieve and to maintain normal penis erection in the
presence of sexual stimulation. Sildenafil is a selective inhibitor of
cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5
(PDE5) enzyme.
How does Cenforce works?
Physiology of penis erection defends upon the
concentration of cGMP in presence of sexual stimulation. Cenforce being a
cGMP specific PDE5 inhibitor promotes the activity of local nitric
oxide by specifically inhibiting PDE5 enzyme (responsible for
biodegradation of cGMP) in corpus cavernosum. Increase level of nitric
acid and subsequently cGMP result in smooth muscle dilatation and
increased influx of blood to the corpus cavernosum.
Cenforce will only exhibit its effect in presence of sexual stimuli.
What are the active ingredients of Cenforce?
Each film coated tablet of Cenforce consists of:
Sildenafil 100 mg
Excipients QS
Coloring agents
Contraindications associated with Cenforce
- It is contraindicated in patients with known history of allergic
hypersensitivity to Sildenafil or any of the other ingredients present
in Cenforce.
- Use of sildenafil and nitroglycerine or nitrate drugs is highly
contraindicated. Sildenafil promotes effect of vasodilator nitric oxide.
So coadministration of sildenafil and nitrate like drugs causes
amplified hypotension effect.
- Cenforce is not recommended in individuals < 18 years and in women
- In patients with severe hepatic and renal dysfunctioning (CLcr <30 mL/min) use of Cenforce is contraindicated.
- Sildenafil is absolutely contraindicated in patient with history
(past 6 months) of ischemic heart disease, myocardial infarction, in
patients with permanent pacemaker, arrhythmia, heart stroke or optic
neuropathy.
What are the warnings/precautions?
- It is recommended to consult physician prior to administration of Cenforce in case of any underlying health condition.
- Dose modification is required in concurrent use of Potent CYP3A4
inhibitors (ketoconazole, ritonavir, atazanavir, clarithromycin,
indinavir, itraconazole, nefazodone) and Cenforce. Since CYP3A4
inhibitors are potentially increase the plasma concentration of
sildenafil.
- Significant alcohol consumption with this medication can cause
symptoms of orthostatic hypotension and syncope like light-headedness,
fainting, weakness, breathlessness, vision and blurring.
- Sexual activity in patients with significant cardiac abnormalities
is associated with potential cardiac risk. So in these patients use of
sildenafil should be carried out under doctor’s supervision.
- Caution should be exercised in concomitant use of antihypertensive
(alpha blockers) and Cenforce may result in additional drop in blood
pressure. It may lead to symptomatic hypotension.
- Patients with penis deformation or suffering from conditions like
sickle cell anaemia, multiple myeloma should be cautious while using
sildenafil.
- Caution is advised while using Sildenafil in patients with bleeding disorders like peptic ulcer
- Risk benefit ration should be assessed while using Sildenafil in patients with untreated diabetic retinopathy.
What are the expected side-effects?
- Painful and prolonged erection
- Headache, dizziness
- Flushing
- Back and limbic pain
- Nasal congestion, rhinitis
- Visual deformity or blurred vision
- Sudden decrease in hearing ability, ringing sound
- Body rashes
- Abdominal discomfort e.g., diarrhoea and gastric bleeding, nausea and vomiting

What are the possible drug interactions?
- Potent CYP3A4 inhibitors and CYP3A4 inducer
- Concurrent use of Cenforce with nitrates like isosorbide and monosorbide nitrate, nitroglycerine
- Antihypertensive agents
- Alcohol
What should be done if dose is missed?
If you missed a dose of Cenforce dose, and you
remember it the same day take it as soon as you remember. But if you
don’t remember until the next day, then never try to compensate a missed
dose. Skip a missed dose and take the drug as it was schedule by your
doctor.
What should be done in case of overdosing?
Symptoms can vary from moderate to severe
gastrointestinal irritation to low blood pressure, tremor and dizziness.
In case of overdosing immediately contact doctor.
What is the Dosage regimen, and how medicine should be taken?
- Cenforce is intended for only oral use.
- This medicine is recommended once daily about 30 minutes before initiation of sexual activity.
- It should we swallowed with water.
- It can be taken with or without food.
- Dose adjustment by physician is required in specific patients’ population.
What are the storage instructions?
* Sildenafil should be stored in cool and dry place, below 20o C temperature.
* Do not expose the medication to extreme heat or freezing temperature.
* Keep out of the reach of children and pets.
* The medicine should be checked properly for the expiry date before applying it.
* Do not flush the medicine if not instructed to do so.
