Make sure you unplug everything, including the USB cable from your computer, to make sure there is no power supplied to the system.

If that doesn't wake the machine, best ping sup...@makerbot.com directly for this one.

Contact Makerbot support.  This is likely a known hardware issue.  But known or unknown, they're going to be able to assist you better.

Is the light on the external power supply lit? If it is unplug it from
the Replicator and plug it back in again. Does it work now? If not
wait for Makerbot.

Take the panel off covering the Motherboard, and see if the 5V
regulator has blown.
