File as input to the program

The Readme file says:

"Also a file can be created with smtp server, from, to, cc, bcc, subject
and mail body which can be used as a single input to the prgram. For
example, if you create a file like below:"

But then it says:

"Note: No Cc, Bcc or Subject can be specified in this file. Use the
appropriate flags if you want to use them."

Can I use cc, bcc and subject in this file of not? How does mailsend
know that it is the Subject line in this file?

The readme file is outdated.

The file may only contain the message text. You can supply some addresses with the "-l" file, the rest has to go on the command line.
