[Lhs-biohacking] Self-cloning workshop

Hi,

I checked the protocol for the self-cloning workshop. The materials used in the kit are easily found and I could provide for free both plasmid and bacteria if we want.
We may still want to buy once the kit just to see how they sell this type of product and then copy them :P

Great. The kit is £100 for 16 'students'. I've emailed Asa from Madlab about whether they bought the whole kit or substituted their won parts.

So what are the parts from the kit that we don't already have? I can see:

  • E. coli and plasmid (Tom can provide)
  • Transformation buffer (random website says this can be made up of 50 mM Tris, 0.35 M sucrose, 0.38 M NaCl, 1 mM MgCl2, 0.00001 % rezasurin)
  • LB broth (£66 from sigma)
  • Recovery buffer
  • Agar + additives (£42 from sigma)

Anything else? So it seems the kit is the way to go unless we can get more of the reagents much cheaper than I found them or for free from Tom / UCL. Asa says they used the standard kit, and that it was very good.

NCBE sell replacement E. coli and plasmids for the kit, implying there'll be enough reagents left over to do it again - and Tom can provide these for free.

I think probably it is better to see how the proper kit works and then to produce our own, it's not so expensive.
I can still provide materials afterwards (or at the same time and compare the two things) but they'll be limited to E.coli and plasmid.
