Hi all;
actually The question is on the title...after looking at the methods on the draft manuscript, I do not understand how could we make sure that we are not losing amplified fragments during library preparation because it is on a "hairpin" configuration?

Hi Marco,
I am not sure I understand the hairpin problem, would you mind explaining in more detail? The LAMP products are palindromic, but as they are double-stranded I don't expect them to form a hairpin. The final PCR amplicon is neither palindromic, nor contains a hairpin. Am I missing something obvious?
