Constraints on Initial Solution/Routes?

Are HardActivityConstraint and HardRouteConstraint are applied to initial solution/routes as well?

What if someone is applying some constraint on a job/service which is in the intial route, but that job never gets ruined and hence never checked for constraints as constraints are applied only when some ruined job is inserted back in the initial solution.

Please clarify this doubt, I am stuck at some point from 2-3 days.
