Most cheats work on the modern control scheme, but you do need to switch to classic for a few of them. It also seems that the speed at which you have to type in the cheats has been increased from the old game, so if a cheat doesn't work, give it at least five tries, and try to memorize it.
I haven't found anyone else who's posted about it, so I thought I might as well do it, so something relevant actually pops up when trying to google "gta san andreas definitive edition cheats not working".
I decided to mess around with some cheats and quickly found that the infinite health cheat doesn't work. Anyone else having this "issue"? I loved that cheat from back in the day it just made free roaming so much more enjoyable just to mess around it'll suck if they intentionally removed it.
Many of the optional cheats from the original GTA: San Andreas return in the Definitive Edition. There are some, such as some of the codes that change all pedestrians, as well as certain weather cheats, that do not work in the remaster due to technical limitations. Thus, it's likely that infinite health was one of those cheat codes. Anybody who has followed GTA Trilogy news knows that the game does have some technical problems. It wouldn't be unheard of if some cheat codes caused issues somewhere in the game.
Infinite health is the main cheat code to not work in GTA San Andreas Definitive Edition. By comparison, the above YouTube video displays all of the cheat codes that still work in GTA San Andreas Definitive Edition. Thus, it's not as if the game got rid of every iconic cheat.
So it would seem as though most cheat codes have returned, but the coveted infinite health one did not. It's a shame for those who enjoyed abusing that specific cheat, as there isn't much in the way of alternatives in GTA San Andreas Definitive Edition.
The only other health-related cheat code is the one that gives players $250K, Health, and Armor. Thankfully for GTA San Andreas Definitive Edition players, it still works. It isn't a full-on replacement for infinite health, but it can heal players if they've taken some damage beforehand.
But some missions are very difficult even with cheats. For example, Robbing Uncle Sam. Though I have ensured CJ's health using a cheat, I have to take care of Ryder's life by loading all 6 boxes before he dies in gun battle with soldiers. Can anyone here tell me how cheats can be of help in such a situation? Or which cheat(s) should I use to pass such a mission?
Yes...! But I have to do it before Ryder dies right? So thats exactly the problem... I can't put all the boxes in time, inside the truck.. So I was wondering whether I could use any cheats to assist. Unfortunately most of the cheats are not of much use in passing missions.

Make sure you select the right code for the cheat you want to use, depending on whether you have the original version or the definitive (trilogy) edition of GTA San Andreas, as the button combinations are slightly different between the two.
Big Head Mode: UP, UP, DOWN, DOWN, LEFT, RIGHT, LEFT, RIGHT, B, A
This is a brand new cheat code only available in the Definitive Edition. It will give all NPCs, as well as CJ, big heads. Does not work in the original version of the game.
Lock Wanted Level: B, RIGHT, B, RIGHT, LEFT, X, Y, UP
Lock your wanted level on whatever number of stars you are. Even though there is a single cheat code, this cheat still works in both the original and Definitive Edition.
Six-Star Wanted Level: B, RIGHT, B, RIGHT, LEFT, X, A, DOWN
Instantly get a 6-star wanted level. Even though there is a single cheat code, this cheat still works in both the original and Definitive Edition.
Maximum Fat: Y, UP, UP, LEFT, RIGHT, X, B, DOWN
CJ gets as fat as possible. Even though there is a single cheat code, this cheat still works in both the original and Definitive Edition.
Maximum Muscle: Y, UP, UP, LEFT, RIGHT, X, B, LEFT
CJ bulks up, obtaining maximum muscle. Even though there is a single cheat code, this cheat still works in both the original and Definitive Edition.
Minimum Fat and Muscle: Y, UP, UP, LEFT, RIGHT, X, B, RIGHT
CJ loses weight and muscle. Even though there is a single cheat code, this cheat still works in both the original and Definitive Edition.
