Joomla mail function problem

I'm trying to change the mail function for a client. Whenever I try to enter the new information, I get the following message upon hitting "apply."
"An Error has occurred! Unable to open configuration.php file to write!"

Where do I look for this file? What is the likely problem?

Permission issue in configuration.php - it is not the right value - you need it to be 640 or higher
