Whole IgG antibodies are isolated as intact molecules from antisera by immunoaffinity chromatography. They have an Fc portion and two antigen binding Fab portions joined together by disulfide bonds and therefore they are divalent. The average molecular weight is reported to be about 160 kDa. The whole IgG form of antibodies is suitable for the majority of immunodetection procedures and is the most cost effective.

Storage and Rehydration: Store freeze-dried solid at 2-8C. Rehydrate with the indicated volume of dH2O (see product specification sheet) and centrifuge if not clear. Prepare working dilution on day of use. Product is stable for about 6 weeks at 2-8C as an undiluted liquid.

Extended Storage after Rehydration: Aliquot and freeze at -70C or below. Avoid repeated freezing and thawing. Alternatively, add an equal volume of glycerol (ACS grade or better) for a final concentration of 50%, and store at -20C as a liquid.

Expiration date: one year from date of rehydration. The expiration date may be extended if test results are acceptable for the intended use.

Horseradish peroxidase (HRP) conjugates are prepared by a modified Nakane and Kawaoi procedure (J. Histochem. Cytochem. 1974. 22, 1084). Peroxidase conjugates are commonly used for immunohistochemistry, Western blotting, and ELISA. Affinity-purified anti-horseradish peroxidase and conjugates are available for detection of horseradish peroxidase antigen or for signal amplification of HRP-containing reagents. For immunostaining of mammalian cells, an advantage of using anti-horseradish peroxidase is reduced background, since the antibody does not recognize the endogenous peroxidase-like enzymes found in those cells.

F(ab')2 fragment antibodies are generated by pepsin digestion of whole IgG antibodies to remove most of the Fc region while leaving some of the hinge region. F(ab')2 fragments have two antigen-binding Fab portions linked together by disulfide bonds and therefore they are divalent. The average molecular weight is about 110 kDa. They are used for specific applications, such as to avoid binding of secondary antibodies to live cells with Fc receptors or to Protein A or Protein G.

1:100 - 1:800 for most applications

Dilution factors are presented in the form of a range because the optimal dilution is a function of many factors, such as antigen density, permeability, etc. The actual dilution used must be determined empirically.

Alexa Fluor 647-conjugated antibodies absorb light maximally around 651 nm and fluoresce maximally around 667 nm. They are brighter than Cy5 and DyLight 650 in aqueous mounting media. Alexa Fluor 647- and APC-conjugated secondary antibodies are the best choice for flow cytometry when secondary antibodies fluorescing at these wavelengths are desired. Alexa Fluor 647 conjugates are the best choice of far red-emitting dyes for multiple-labeling detection with a confocal microscope.

A significant advantage of using Alexa Fluor 647 over lower wavelength-emitting dyes is the low autofluorescence of biological specimens in this region of the spectrum. However, because of its peak emission at 667 nm, Alexa Fluor 647 cannot be seen well by eye, and it cannot be excited optimally with a mercury lamp. Therefore, Alexa Fluor 647 is not recommended for use with conventional epifluorescent microscopes. It is most commonly visualized with a confocal microscope equipped with an appropriate laser for excitation and a far-red detector. Alexa Fluor 647 conjugates are less expensive alternatives to allophycocyanin conjugates for flow cytometry.

I cut and rearranged other four-panel versions of the Midjourney hamsters to create more coherent stories of hamster disaster. Here, a hamster (mouse, really) is surprised by a bright light above his head, which sucks him inside, leaving an empty shed.

Hamsters are protected by The Animal Welfare Act but sadly the basic needs of these creatures have been ignored by the pet industry for too many years. Furthermore, the products they sell often put the health and welfare of hamsters at risk.

Hamsterwelfare.com has been created to expose the injustices that hamsters are experiencing at the hands of the pet industry. The correct animal welfare advice exists but it is constantly ignored by the industry and when unsuitable products are sold to consumers, those pet owners may be unwittingly causing harm to their pet and breaking the law.

Pet shops rarely sell the correct minimum cage size which The Veterinary Association for Animal Welfare (TVT) recommends to be a minimum 100cm x 50cm x 50cm (L x W x H). This is recommended for ALL species of hamsters including dwarf. At best a Pet shop may stock one type of cage that meets the bare minimum size but nowadays there are too many small cages which look more like toys and are targeted to appeal to children rather than adequately meeting the needs of the hamster that has to live inside them. Recommended Cages and Tanks can be seen here.

Hamsters can run 9km a night and their health requires the exercise. Research shows that small wheels cause spinal curvature injuries which can result in permanent unnecessary pain and suffering. The protection that hamsters are given under the Animal Welfare Act should be used to prevent owners from purchasing wheels that can cause pain and suffering. Research shows that Syrian hamsters require a wheel size of at least 30cm (The community commonly accept 28cm as being suitable) and dwarf hamsters 20cm.

To the frustration of many Hamster owners the wrong labelling of wheel size and suitability is common practice in the pet industry. Small wheels are often labelled as suitable for both Dwarf and Syrian Hamsters.

This advice has been ignored for many years and some manufacturers even go as far as saying that their product is safe! There are many testimonials from the public saying that fluffy bedding has caused injury or death to their pet.

One theory is that the small fibres stick to the inside of a hamsters cheeks and then becomes ingested. Of course the ingestion issue is impossible to see with the naked eye but new ground breaking evidence has come to surface.

The combination of this evidence and advice from the most reputable sources such as The RSPCA, The Blue Cross and Royal Veterinary College, many testimonials over the years and public outcry involving a Change.org petition, has persuaded many major retailers to stop putting animals in harms way.

"Cute", "pretty", "weird" and "fast" are just some of the adjectives used to describe the Roborovski hamster and these words describe this different hamster perfectly. I myself think that the Roborovski is one of the prettiest and most fascinating hamsters. Their style and relationship with other hamsters is unlike any other dwarf hamster.

The Roborovski, phodopus roborovskii, came into the UK in May 1990. Unlike other dwarf hamsters the Roborovski does not have the black dorsal stripe. They have slightly longer legs than other dwarf hamsters yet overall size when fully grown is 4-5cm rather than 7cm. Their top coat colour is dark chestnut/gold with a slate grey undercolour. The belly fur and side arches are white. They have white eyebrows and the ears are more noticeable than on other dwarf hamsters and they have black beady eyes. The roborovski is not a pet to pick up and stroke. Due to their slightly longer legs they can run very quickly (in fact they can shift!) and if they put their minds to it they can jump up to 2 feet in height. Believe me, trying to catch a Roborovski hamster on a beige carpet is murder!

It is best to keep Roborovski hamsters in a large glass or plastic tank covered with wire mesh. They are very active and so require lots of toys to play with - and they do play. They love to chew, jump, climb, hang upside down etc and they love to do this in groups. I must admit that watching a group of Roborovski hamsters can be more fun than watching TV! Mating can be a real headache - they are not as prolific as other dwarfs but once they start they continue for four litters with an average of six babies in each litter. The gestation period is 26-28 days and for the male to catch the female to mate with her is quite a feat because they are on the move the whole time. Once the female has been mated and produces her litter, every hamster helps out. The babies are not allowed to be left on their own, with one hamster baby-sitting while mum pops out. For the first 2 weeks development is slow but once out of the nest and eating solid food thebabies develop quickly and by now mum is pregnant again.
