MW correction of STA particles

Hello!

I know this isn't the standard workflow, but I'm interested in performing missing wedge correction and denoising on subtomograms that have been aligned using STA. Unbinned, the dataset is ~6k particles at 1.36A/pix and a 256pix box size. 

Have others tried this and observed improvements in anisotropy? I've been experimenting with it but don't usually see any. Advice on parameter selection would be most helpful!

Hi Micah,

First of all, what you propose is, as far as I understand, exactly what is shown in panels d,e of the IsoNet paper Fig 1, so it should be possible:

Alternatively, how about running IsoNet on the tomograms (as usual), then extracting and averaging the subvolumes from the MW-corrected tomos using your existing alignment parameters? Unclear how well this will work in practice, but might be worth a shot?
