Tilt correction

Hi,

Should we correct the tilt angle offset before running IsoNet, or will this create problems? The correction certainly rotates the missing wedge away from the Z-axis. Thanks.

Hi Reza,

It would be better to make the tilt angle symmetric. For example, -60 to 60 rather than -50-70. To achieve this, typically set tilt angle offset to 0 for non-FIB samples.  

While using a different tilt angle offset won't cause errors, a tilt scheme deviating from what was modeled in IsoNet may result in suboptimal outcomes.

Hi,

Thank you for the help. I may have used the wrong terminology. I'm referring to the fact that my tomograms are tilted with respect to the Z-axis. Here is a picture from the AreTomo 1.3.0 manual for correcting the tilt angle offset:

Hi Reza,

I think what you can do is to check the final alignment file ".aln" or ".tlt". If the final aligned positive tilt angle is roughly equal to the positive tilt angle, it is good for IsoNet. Otherwise, you can adjust the offset. 

You can also try IsoNet with both with and without the correction.
