Applying IsoNet with other filters

Dear Dr. Liu,

As a newbie in cryo-EM, I have a conceptual question.

I would like to  compensate the missing wedge of my tomograms, with IsoNet, and denoise them with other apps such as tomo3d and topaz-denoise3D.

The filters in Tomo3d works with tiltseries during the reconstruction process. Therefore, I have to apply Tomo3D to my data before proceeding to IsoNet. My questions is, is IsoNet still functional if I train it after applying tomo3d filters? Does it make sense of treating my data with multiple software?

Dear Chris,

The answer is yes, it's okay to treat your data with multiple software before Isonet.
And Isonet works better when your input tomograms have higher contrast. for example: Isonet deconv step is made for the purpose to increase contrast.
