What you can do right now is create a desktop shortcut for ImageGlass, then modify its properties to add your desired "default directory". When you double-click the shortcut, ImageGlass will run and show images from that folder.
E.g. assuming your default directory is "E:\My pictures", you can set up your desktop shortcut like so:
