Spot Detector Bug (either from Spot detector OR from me)

Hi Happy list Members,
We are trying to detect spots in Yeast cells. The spot detector circles the spots we want but the digital image doesn't correspond
 (see enclosed image) . The xls output gives the size of the binary image areas and not of the detected spots .
Are we doing something wrong ?

Hi Eric,
it's nice to see that you are already using Icy just 2 days after the Inserm formation. ;o)
Your picture seems to correspond to a basal intensity threshold and not to the spot detector analysis (shape of the cell).
Did you take the picture inside the save folder ?

Yes I took the binary image from the save folder in which I also found the xls file!!!!

Hi Eric,
after having a detailed look on your picture, the binary seems to be indeed ok. Actually the data you get in the excel files contains the connected components corresponding to your selected parameters (scale 2, 60). The 4th components correspond to the plasma membrane (surface 661, with the mass center on the top of the cell (neer the budding, see detection index 4 on capture 13.21.56). If you want to avoid plasma membrane detection, you can filter your results based on the size - for exemple 1-100 (see capture 13.24.18). Then you will have a binary and a xls files with only your "little spots".
I send you a zip file, with the filered and not filtered version of the spot detector.

Hi IcyLyd,
I finally understood that Spot detector is a frequency filter and not a spot detector (am I wrong ???). My membrane being thin (in the same "scale" than my vesicles) it is detected by the frequency filter !!!!
