Hi SyedMuhd,

To turn on Vibrate for a specific application, open the Application Preferences and change the Default Display property to Vibrate. If you want sound and vibration, choose Growl and Vibrate.

To bring up the Application Preferences, you can tap on an existing notification in the history list on the main screen and choose the first menu item "App:".
Alternatively, you can press the Menu button on the main screen, then choose Applications, then tap the application that you wish to change.

Thanks,
  Carey

No, unfortunately there's no ability to use a custom notification sound in the current version.

Thanks,
