On my system, launching the app again from the Apps folder or Spotlight brings up the prefs pane. Try quitting the app from the Activity Monitor, then launch it fresh and see if that brings up the prefs pane.

Also, from the Growl notification that pops up when selecting No Icon:

Enabling this option will cause Growl to run without showing a dock icon or a menu item.

To access preferences, tap Growl in Launchpad, or open Growl in Finder.

... so it should work.

Thanks for these replies. Clicking on the Growl icon in Launchpad didn't do anything. But launching from the Apps folder did, and to make things better / more confusing, going from Launchpad behaves properly now.

If Growl is in the background and you try to launch it again in any way (finder, spotlight, launchpad) then the preferences will show up. This is our solution so that people can run Growl in the background but still get into preferences.
