That's a bug on apple's side that we're chasing down and so far we don't have a good answer. Can you check for logs in Console to see if there is anything useful from Growl when you do that?

Growl is no longer a preferencePane, you'll want to perform the following steps to get back up and running again.

kill GrowlHelperApp via Activity Monitor
open System Preferences
right click on the Growl entry
choose remove "Growl" Preference Pane
launch MacAppStore.app
click the purchases tab
click the install button next to Growl
open /Applications
locate Growl.app
launch it
