Are there any API available to create/update the custom  Creative templates .

And create the list Variables inside it .

Based on the documentation, you can utilize the CreateCreativesFromTemplates.java code example to generate a new template creative for a specified advertiser. To identify existing creative templates, you may execute the GetAllCreativeTemplates.java. It's essential to note that directly creating a creative template through the API isn't supported. However, by using the already existing creative templates, you can successfully generate new creatives using the API.

I can confirm that the provided sample code is the only available resource for creating creatives using the creative templates via the API. You may refer to this code sample to create a custom creative but not the custom creative template via the API.
