You'll want to convert your shapefile into an AOI, then create a signature
file from your AOI and your image.
1) Open shapefile on your image and display as polygon
2) Select all polygons
3) From viewer AOI pull-down menu, select copy selection to AOI. Save this
AOI file. Select all AOIs.
4) From Signature Editor, hit the third button from left to "Create New
Signature(s) from AOI".
