Hi,

I am new to using Spring.

I used e2helixboxer to box all the helices I want to use (*boxes.txt files).

However, Spring ran into en error of 'spring.db' not found in the newly created directory when I ran segment in spring2D.

What did I do wrong? I ticked CFTTILT.

Thanks,

Ming

Dear Ming,

You will have to run MICCTFDETERMINE and provide the spring.db from that run. This database contains all the required information about CTF parameters to perform correction on the segments.

Best wishes,

Carsten

Hi Carsten:

Thanks for your answer. I did run MICCTFDETERMINE and see the spring.db file in the directory generated by MICCTFDETERMINE.

However, I do not know how to let segment find that file. When I run segment it generates a new directory and could not find spring.db in the new directory.

I found the spring.db file option Segment is running now.

Thanks,

Ming
