Introduction:
Ori and the Blind Forest: Definitive Edition is new version of the metroidvania from 2015 that many people had a Game of the Year contender and/or winner. This version fixed a number of small issues, added an entire new area, new abilities, fast travel via Spirit Wells, a new Easy difficulty, and a number of other little tweaks. It really is a definitive version of an already outstanding game.
Step 1: Hard Run
The first playthrough will be to simply complete the game on Hard. Don't worry about deaths or time, and don't go too out of your way for any collectibles. Doing the miscellaneous combat-related achievements might be a good idea though.
Step 2: No Abilities Speed Run - Then 100%
Doing these in their own run is helpful because you will want the extra help staying alive during your no death run that the abilities in the Efficiency tree provides, as well as extra combat skills to quickly take out enemies you can not avoid. The speed portion will come along since you won't be going out of your way for extra experience orbs or anything else, and since the game now has fast travel, it goes even faster. You can attempt to combine this with Step 3 if you'd like, but it is no easy task. If you do these on their own, play on the new Easy difficulty.
Once you've beat the game and unlocked those two achievements, load your game back up and go for 100% completion. Collect everything and uncover the entire map, as well as max out your abilities. It is better to do this here rather than on Hard, because Hard difficulty requires many more skill points to unlock the later abilities, which will leave you grinding kills for a long time.

Step 3: One Life Difficulty
Just like in the original version of Ori and its "Immortal" achievement, you can abuse the save system and playing on/offline to allow yourself a relatively easy way to complete this achievement, which is described in more detail in the "Unhinged" achievement.
Step 4: Mop Up
You shouldn't have any achievements left at this point, but if for some reason you need more kills or never did a specific combat-related achievement, refer to those now to finish up your 1250.
