Is it possible to mount to ISOs at the same time?

I want to mount 2 ISOs at the same time. or can i create persistence linux iso using single iso?

If you want 2 CD drives to show up, you'll need a kernel that supports multiple host devices.
If the kernel you have now doesn't support this, you probably need to delve into the code of your kernel to compile one yourself.

Can you give few example kernels/Roms which support multiple host devices.

Some phones that have a SD-card slot will support multiple host devices, since they use that to host the SD-card to a PC. Of the phones I have, the Moto G 2015 and Galaxy S i9000 both have 2 of them. Since most custom roms are usually based on these stock kernels, they'll all support 2 hosting devices on these phones.
If you have a phone that comes with 1 hosting device, chances are that no other rom will have multiple hosting devices.

I'm trying using OnePlus one. If the phone supports multiple host devices, how can i mount 2 images?

Ah, the OnePlus One only has one hosting device. You'll have to patch and compile your kernel to get multiple hosting devices.
Once your kernel has multiple hosting devices, you'll get more options in the hosting dialog that pops up when you tap an image in DriveDroid. You can see an example here: http://softwarebakery.com/boot-windows-from-drivedroid/drivedroid-03.png
That way you can host one image on one hosting device and another image on another.

I made 2 partitions in a single img file, but one is mounting and the other is not mounting (showing error). Is it possible to mount more than one partition and is there any limitation for type of file system?

Yes, this is certainly possible. There must be some other reason why the second partition is not mounting. The USB device just acts like any other USB device: a big block of bytes. What partition table you put on it and which filesystems you format the partitions is up to you.
