Hi guys, wondering if anyone has the pRS413 yeast artificial chromosome on stock and would be able to share. Will paypal the money for shipment and handling

Another YAC with LoxP would also be cool, but I don't think they made many of these

Hey Andreas,

Our lab has pRS413, but it's a plasmid and not a YAC based on the Addgene entry associated with it:

We also have yeast-compatible plasmids containing loxP sites. If you want the plasmids, I can see if our material transfer agreement allows us to share them. If so, I can ship you a vial containing enough plasmid to transform into E coli for you to generate your own stock.

Let me know!

Best,
Sean

That would be great,

As far as I understand pRS413 and similar plasmids with Cen/Ars are kind of hybrid self-replicating plasmoids (single copy, contain centromere) that can replicate 200 kbp. That would qualify them as YAC for me ;)

Would appreciate you checking your MTA, but if shearing isn't possible, I'm sure I can find another source.

What other loxP YACs do you have?
