OpenTAQ: A Review

I don't know if anyone else has posted a review of opentaq yet, so I wanted to put a comment out about it.

I ordered the Taq via express mail for $40/100 rxns +$23 shipping.  It was shipped in an overnight envelope at room temperature in a bag with a data/useage sheet.  The datasheet was easy to read and well laid out. Due to a mail snafu it did not reach me until 2 days after shipping- when i immediately put it on ice, and took it to the lab.

Unfortunately, I had forgotten my primers, but some people at the pcr hackathon whipped up some test dna with a plate that had been contaminated by fungus, and some draino.  The DNA was run against two fungal barcoding primers which we had on hand: ITS1 and TS4, although I don't remember which went where.

  1. negative control     w/ Opentaq
  2. Barcoding primer    w/Opentaq
  3. Barcoding primer 2 w/Opentaq
  4. DNA ladder
  5. negative control
  6. Barcoding primer    w/Neb 2x
  7. Barcoding primer 2 w/Neb 2x
  8. Barcoding primer 2 w/Neb 2x

The NEB and Opentaq samples were run on different machines, but this gel shows good amplification using the opentaq master mix, even after it was left out at room temp for a day+

I am impressed and happy with the product.  My only criticism is that it would be nice if you got your tracking # with your order.

I was impatient and ran it at like 160V and 60-70? mA and the first four lanes (neg control, opentaq, opentaq, ladder) were loaded first and had a lot more time to settle and diffuse. As you can see, the ladder also streaked, so we are not sure what it is, but we think it has something to do with either the voltage/current settings and a lot of DNA, or the loading.

I want to stress that this is not meant to be a comparison between the two enzymes, as they were run under completely different conditions- just thought people might be curious.

Thanks the the review. We'd like to try this, and I've been meaning to find out how to get it shipped to the UK.

You can get this taq, or the plasmid and protocol to make it at http://www.openbiotech.com/

I would advise contacting the openbiotech folks directly about shipping to you.  Their contact info is in a diybio googlegroup thread somewhere about openbiotech/ opentaq but I dont have it on hand.  I remember that they replied to the thread though.
