Hello Everyone,

I am excited to announce that we can officially be considered a Giardia lab.  Cells are growing and we did our first transformation today.  I want to thank the Cande Lab for answering all our annoying questions (especially Stephane) and the Dawson Lab (especially  Kari) for helping us get going.

 

I also have a question.  We have been using Difco Bacto Yeast Extract.  There are other brands at half the cost.  Does anyone use another brand of yeast extract?  This is not huge money but it could add up over the next 30 years.

We are using BD's Bacto yeast extract. Ref number is 212750. I am not sure it is the same as DIFCO's brand.

Yes I meant Bacto, we are using the same thing as you 212750.  Bacto and Difco are both BD brands hence my confusion.  Has anyone tried non BD yeast extract? We were told to use ascorbic acid and cysteine from Sigma but the Cande lab has been using stuff from Fisher without any issues.  I am just wondering what is so special about the Bacto brand?

Hey- alex--WELCOME AND CONGRATS!!!

excystation is VERy FINICKY and so whenever we needed new serum, YE or casein, we would test all available large lots--to see which the parasites liked the best--we screened first for growth, then encystation and excystation and bought a huge amount of the best--we thought it minimized variation.

The most finicky part of the media is cysteine and we tended to get it in small bottles and keep it dessicated to minimize auto-oxidation--often if we had problems with media it was cysteine.

A lot is "habit."

We used Difco Bacto before. Now we use yeast extract from Fluka (70161). It works fine. Our tryptone (T9410) is also from Fluka.  You might try those.

It's great to hear that your lab is up and running, Alex!  We are still using BD 212750.  We did run out of it one day and bought EMD 1.03753.0500 granulated yeast extract from our on-campus store as a temporary replacement. I wouldn't recommend that one. It seemed to be a very different product, and because it was granulated, it didn't dissolve well. We ended up borrowing the Bacto yeast extract from another lab instead--it's widely used in the microbiology labs here.
