In this study, we have characterized IBS004735 that was obtained in the same screening. IBS004735 also promotes myogenesis in C2C12 cells, but is not similar to IBS008738 in the structure. IBS004735 activates TAZ via Akt and has no effect on TAZ phosphorylation, which is the well-described key modification to regulate TAZ activity. Thus, we introduce IBS004735 as a novel TAZ activator that regulates TAZ in a yet unidentified mechanism.

If binding to the activator can be restored, for examples by a small molecule, this may represent the drug needed to cure the disease. Here, a single-chain antibody fragment, or nanobody, called VHH-BS2, which specifically binds TorsinA and LULL1 has been found. Using this nanobody as a crystallization chaperone, both TorsinA-LULL1-VHH-BS2 and mutant TorsinADE302/303-LULL1-VHH-BS2 complexes were crystallized and the structures solved at very high resolution (1.4 Å). Pretreatment of JB6 P+ mouse epidermal cells with lingonberry extracts produced a dose-dependent inhibition of activator protein-1 (AP-1) and nuclear factor-kappaB (NF-'B) induced by either 12-O-tetradecanoylphorbol-13-acetate (TPA) or ultraviolet-B (UVB) light. Lingonberry extracts blocked UVB-induced phosphorylation of the mitogen-activated protein kinase (MAPK) family members ERK1, ERK2, and p38 but not JNK. Lingonberry extracts also prevented TPA-induced phosphorylation of ERK1 and ERK2. Results of soft agar assays indicated that lingonberry extracts suppressed TPA-induced neoplastic transformation of JB6 P+ cells in a dose-dependent manner. Lingonberry extracts also induced the apoptosis of human leukemia HL-60 cells in a dose-independent manner. These results suggest that ERK1 and ERK2 may be inhibited by lingonberries, which results in suppression of AP-1 and neoplastic transformation in JB6 P+ cells and causes cancer cell death by an apoptotic mechanism in human leukemia HL-60 cells.
