Physical evidence is any tangible object that can connect an person to a crime scene. Biological evidence, which contains DNA, is a type of physical evidence. However, biological evidence is not always visible to the naked eye. DNA testing has expanded the types of useful biological evidence. All biological evidence found at crime scenes can be subjected to DNA testing. Samples such as feces and vomit can be tested, but may not be routinely accepted by laboratories for testing.
Since only a few cells can be sufficient to obtain useful DNA information to help your case, the list below identifies some common items of evidence that you may need to collect, the possible location of the DNA on the evidence, and the biological source containing the cells. Remember that just because you cannot see a stain does not mean there are not enough cells for DNA typing. Further, DNA does more than just identify the source of the sample; it can place a known individual at a crime scene, in a home, or in a room where the suspect claimed not to have been. It can refute a claim of self-defense and put a weapon in the suspect's hand. It can change a story from an alibi to one of consent. The more officers know how to use DNA, the more powerful a tool it becomes

In addition, if laboratory analysis reveals that DNA evidence was contaminated, it may be necessary to identify persons who have handled that evidence.
The first responding officer may be called upon to transport evidence from a crime scene. As with any evidence, the officer should ensure that the chain of custody is maintained. In addition, they should be aware that direct sunlight and warmer conditions may degrade DNA, and avoid storing evidence in places that may get hot, such as the trunk of the police car. To best preserve DNA evidence, store in a cold environment.

There is some controversy as to when some of the churches were constructed. David Buxton established the generally accepted chronology, noting that "two of them follow, with great fidelity of detail, the tradition represented by Debra Damo as modified at Yemrahana Kristos."[15] Since the time spent to carve these structures from the living rock must have taken longer than the few decades of King Lalibela's reign, Buxton assumes that the work extended into the 14th century.[16] However, David Phillipson, professor of African archeology at University of Cambridge, has proposed that the churches of Merkorios, Gabriel-Rufael, and Danagel were initially carved out of the rock half a millennium earlier, as fortifications or other palace structures in the waning days of the Kingdom of Aksum, and that Lalibela's name simply came to be associated with them after his death.[17] On the other hand, local historian Getachew Mekonnen credits Gebre Mesqel Lalibela, Lalibela's queen, with having one of the rock-hewn churches, Biete Abba Libanos, built as a memorial for her husband after his death.[18]
Contrary to claims made by pseudoarchaeologist writers like Graham Hancock, Buxton states the great rock-hewn churches of Lalibela were not built with the help of the Knights Templar; asserting abundant evidence exists to show that they were produced solely by medieval Ethiopian civilization. For example, while Buxton notes the existence of a tradition that "Abyssinians invoked the aid of foreigners" to construct these monolithic churches, and admits that "there are clearly signs of Coptic influence in some decorative details" (hardly surprising given the theological, ecclesiastical, and cultural links between the Orthodox Tewahedo and Coptic Orthodox Churches), he is adamant about the native origins of these creations: "But the significant fact remains that the rock-churches continue to follow the style of the local built-up prototypes, which themselves retain clear evidence of their basically Axumite origin."[19]

Well here at the opposite end of the UK, in sunny Sussex, the warm weather seems to have started a mega flow from the bramble such that a well known supplier had run out of wired foundation earlier this week. I have now used all my supers and we are still in June. And only today a swarm that I collected six weeks ago,and then united with a nuc a couple of weeks ago, has swarmed again, on open queen cells. Just when I thought everything was under control!

While it seems true that bees without a queen excluder tend to go up sooner, there may be a price to pay. Without an excluder, the bees sometimes neglect to fill the outer frames in the brood box, say those in positions 1 and 10, and possibly 2 and 9. Instead they work above the established brood nest, which is generally on the center frames.
But getting back to the queen excluder, this situation (honey above but none below) is more likely to happen when the beekeeper has encouraged his bees to build in the supers before they are ready. The bees would have been better off if the beekeeper left them alone.
But by the time that is done, the summer solstice is near and egg laying gradually slows and the populations begin to drop. As the brood nest shrinks the bees backfill much of that with honey to get ready for the winter. This is exactly the time that new beekeepers put on their first honey supers. When it becomes obvious that their bees are not interested in this new space, the beekeeper becomes completely discouraged and blames the queen excluder.
I use excluders a lot, but only for the benefit of housing two queens in the same hive. I find excluders very dangerous in the spring, since they can get the bees into a swarming state very quickly. Also, it kind of goes against the Checker Boarding manipulation. In the summer, after the swarm cutoff date, I think the excluders can be used pretty safely.
I never use the excluder, as my bees and queen roam across a never ending brood chamber, which consists of 3 deeps. Any surplus is above these deeps, but only after the deeps are ready for winter. In a good year that is end of June first of July time frame. A bad year, no surplus.
