Thistrick is a good way to fill the lanes with mushrooms quickly for Buff-Shroom, Punish-Shroom, Muscle Sprout, and Astro-Shroom's abilities, or to make profit with transforming cards like Pineclone and Petal-Morphosis. But although many strategies for optimizing this card exist, many require larger amounts of sun or up to three cards, making them strong in theory but in practice rather impractical.

As such, even if this card has good synergy with your deck or you are assembling a combo, it is generally recommended to simply play this card as soon as possible for the best chance of making a meaningful impact on the field without any problems. Do not hold onto this card too long, or you will end up wasting time and losing out on any possible impact.


Be careful when playing against a Beastly hero, especially if you notice they are running multiple Frenzy zombies. Most zombies with Frenzy will simply destroy Button Mushroom, then attack again to hurt you. Vimpire and Ancient Vimpire (for the latter, any Frenzy zombie is also included) in particular are dangerous as they get stronger after destroying the Button Mushroom. Even outside of Frenzy zombies, Immorticia and Brain Freeze have access to Acid Rain, a trick capable of destroying your Button Mushrooms at once.


Against Impfinity, if he doesn't play anything on the Zombies Play phase on turn 1, you can play this straight away to basically discourage a possible Triple Threat coming; most opponents will hold back on playing Triple Threat, a powerful source of early-game impact, since they do not want to risk the Impfinity Clones being made on the same lanes as the Button Mushrooms.


Due to Button Mushroom not having Team-Up, it is a bad idea to use this when there are less than two lanes that are empty. The only exceptions are when you need to block a strong zombie and use the Button Mushrooms for Evolution.


If you go later into the game, this superpower's worth diminishes due to the Button Mushrooms taking up space where your stronger plants could be played instead. Be sure to avoid playing this card if you know you need (or will need) the space to play more impactful plants.


This won't benefit your opponent unless they are going for a rush deck, which will cause either some of your zombies to be low on health or have you take plenty of damage. If the plant hero uses this on the first turn, it is possible that you will have to accept taking 2 damage unless you have a zombie in one of the Button Mushroom's lanes, or tricks such as Bungee Plumber and Nibble to take out at least one of them.


While tricks like those work, Missile Madness will take them both out. You could also use Extinction Event, but it isn't worth using it on Button Mushroom in general, and you will have to wait until the next turn.

When thriving bacterial cells are threatened by harsh conditions and nutritional restrictions, the extreme stress can drive them into survival mode, forming endospores. These bacterial spores can be exceptionally tough, remaining viable even after exposure to extreme conditions due to their resistance to enzymatic lysis, desiccation, radiation, high temperatures, and chemical treatments such as disinfectants and denaturants. These resistances can be very challenging to metagenomic studies, as efficient lysis of spores is important to ensure an accurate representation of the microbial profile of a sample. Inefficient lysis of spores can lead to misrepresentation of the microbial profile and very little to no DNA recovery from the spores.


The extreme hardiness of spores and their resistance to lysis is a product of many factors. The outer most coating of the endospore is a proteinous layer that provides substantial enzymatic and chemical resistance. Beneath this layer are structures that provide further physical and chemical barriers: a peptidoglycan cell wall called the cortex, followed by a germ cell wall, and an inner membrane. Within this inner membrane is the core, which contains the DNA and ribosomes, and includes additional protective elements including dipicolinic acid and proteins that further protect the DNA from radiation and chemical damage.


To determine the efficiency of different lysis mechanisms on spores, Zymo Research compared three methods of lysis on Bacillus subtilis endospores: chemical, mechanical, and heat. The results indicate that the only way to achieve complete, unbiased lysis of bacterial endospores is to use high-density bead-beating beads with high speed or low speed homogenizers.


Bacterial spore formation was induced by inoculating Bacillus subtilis cells growth/sporulating medium with Bacillus subtilis and incubating for several days. Successful bacterial spore formation was determined by using Schaeffer and Fulton Spore Stain Kit (Sigma Aldrich) and viewed by microscopy. Bacterial spores were then concentrated and frozen in a PBS/ glycerol solution. Cell counting was not possible due to the extremely small size of the bacterial spores, so indirect counting was performed via plating serial dilutions of bacterial spore suspension.


Chemical lysis was attempted using several common commercially available lysis buffers (e.g. Zymo Research Genomic Lysis Buffer, Qiagen Buffer AL, and Qiagen Buffer AVL) which led to no significant DNA recovery, as anticipated (data not shown). This does not reflect a comprehensive review of chemicals with the potential to lyse endospores, it is just an evaluation of some of the most used cellular lysis buffers.


Bacillus subtilis spores were treated at room temperature (RT), 55 C, 75 C, and 95 C for 1 hour. In all instances no substantial quantities of DNA were recovered in the purification, however spores treated at 95 C for 1 hour experienced 106 decrease in cell growth when plated, in comparison to other spore treatments (Figure 1).


The ZymoBIOMICS DNA Miniprep was compared to the DNeasy PowerSoil (Qiagen) kit to evaluate the lysis efficiency and recovery of DNA from solutions containing 6.0 x 108 CFU of B. subtilis endospores. The ZymoBIOMICS DNA Miniprep was consistently able to lyse the B. subtilis endospores and recover the DNA, while the DNeasy PowerSoil kit was incapable of recovering measurable quantities of DNA (Figure 3).


My computer's too new to run spore. Or at least the version I had before, although it was super buggy. All stars and planets were called BAD_DATA, so a mission was "Go to planet BAD_DATA, orbiting the star BAD_DATA, which is within 20 parsecs of the red star BAD_DATA."


go to your library, right click on spore, remove local content. then install again. once done with that and updates, log in and you should be good...but you'll have lost any unshared work and all downloaded content


If you like the look of darkspore, I highly recommend downloading and installing the Spore mod, "Dark Injection" which gives the UI a darkspore style, and adds a ton of Darkspore parts to the creature creator.
