I am looking for an advice regarding microscope. Which fluorescent microscope do you use and which one you can recommend for FISH analysis?

Hi Anastasia, there are lead brands, but after several decades we are satisfied with SEIZZ brand, axioscope microscopes. Banded chromosomes and FISH are sharply defined.

Consider the top quality seizz lens as planapochromatic or neofluar. 

FISH filter cube set must be for fluorochromes: 1- GREEN, 2- orange/red, 3- DAPI and 4- triple band pass (common FISH test uses this combination). All of them according to emission and absorbance (excitation) of the proper FISH probes. Consider that if you will use a capture and analysis software, the integrated image will not come from the triple band pass filter, but from the an integrated image from 3 pictures (fre, green, DAPI)

Hi Anastasia,

First of all, it is important which system you will use for which purpose (spot counting, metaphase FISh, tissue FISH, chromosome analysis etc.).
If you are going to use digital imaging and a support software on this microscope, you should choose this, metasystem, cytovision etc. I recommend you to test the demos of these softwares and the performance of the system for your purpose.
Finding the appropriate number of fluorescent wheels for the FISH probes you will use
Whether this system is automatic or manual, depending on your test numbers,
Requirement for an automatic signal counting slide scanning system or manual scanning and analysis,
the cost of upgrading the capacity of the system in the future
etc.
You should decide in many respects and decide on a cost-effective site according to your needs.

Thank you all for your reply.
Currently we do interphase FISH in our lab (on FFPE tissue, blood and bone marrow samples), using mainly ZytoVision probes and analyse slides manualy (using Olympus AX70)
Looking for a new microscope and software (for update).

Hi Anastasia, 

We use Zeiss motorised FISH and light Axioscopes with the Metafer scanner, and analyse Ikaros and ISIS software. They're very good for high throughput laboratories, and the options for FISH are pretty good. Have used these for over 15 years now. Came from manual Olympus BX  series  in the early days of our lab. The motorisation options for objectives and filter turret is excellent to have. The buttons can be a bit troublesome though but Zeiss look after us pretty well when things go wrong. We have 5 of these in the FISH lab.

I need comment regarding karyotype of a 27 yrs old female patient. 

There is a light band on chromosome 3p.

Please comment on how to interpret this.
