What is the most reliable method for calculating the G-band
resolution?

We use the method developed in our lab (Vancouver General Hospital)
and published in AGT's journal:

Evaluation of Chromosome Banding Resolution: A Simple Guide for
Laboratory Quality Assurance, Josifek, Haessig and Pantzar. Applied
Cytogenetics17(4),1991.

It uses areas on several chromosomes to determine the overall banding
resolution - we find it quite accurate.

Can you maybe post a link to this article or maybe a .pdf version of
it. I tried to find it on the internet but I couldnt find it anywhere,
not even on the AGT site.

Hi This website gives you latest banding resolution from the ISCN2009
booklet.

I use chromosome 16 to determine resolution, if the total number of
bands in chromosome 16 is 8 or more bands it is high resolution
banding in my laboratory.

Our laboratory determines G-banding stage
as follows: -

400 Band stage per haploid set, e.g. two distinct bands on each of 8p
and 9p and three distinct dark bands in the middle of 5q (5q14, 5q21
and 5q23)

550 band stage per haploid set, e.g. four distinct bands on each of
18q, 3 on 11p; 7q33 and 7q35 resolve and 22q13.2 will be visible.

850 bands per haploid set are , e.g. 6q16 should split, 6q24, 6q25.2
and 6q26 appear as three distinct bands; 11p14.1 resolves from
11p14.3; 15q12 is distinct; 20p has at least two dark bands.
