You can, as only conditions for prior to INC-20A filing that No borrowings or Business activities 9any business decisions are allowed) either by Board or General meeting which considerd as business in natures.

as Resignation does not required any approval or resolution of board, resignation if effective one can be file form DIR-12 ( if system does not accept any form prior to INC-20A raise ticket)

You need to see if portal allows the same or not. Legally its allowed but practically check the same from MCA portal
