can I transfect the lentiCRISPR v2 with VSV-G and pCMV-dR8.9 packaging plasmid in 293 K cells?

Hi to anyone has the experience using LentiCRISPR v2,
I have questions about the lentiCRISPR v2, 
1) can I transfect the lentiCRISPR v2 with VSV-G and pCMV-dR8.9 packaging plasmid in 293 K cells?
2) do I need the empty vector for negative control?

293 k? Do you mean 293 or 293T?

What is is 293K. I have never heard of this 293 version.

sorry, it is 293T cells.

293T is primarily used for lenti packaging. I've used lentiCRISPR with psPAX2 and pMD2.G to transfect 293T and make virus.

Hi Hongzhi,

We have put together a screening protocol that includes lentiviral production that you might find helpful here. I have also recently found that Lipofectamine 3000 is a better alternative to lipofectamine 2000, and am attaching the updated protocol using lipofectamine 3000.
