Has Anyone Successfully Used pMBP-CRISPR-Cas12a? Looking for Advice

Hi all,

My name is Eduarda, and I’m a PhD student in Chemical Engineering. I’m working on a project involving CRISPR-Cas12a, and I’ve been trying to express the protein using the pMBP-CRISPR-Cas12a plasmid (Addgene #113431).

Unfortunately, despite several attempts and multiple troubleshooting strategies, I haven’t been successful so far. I was wondering if anyone with experience using this plasmid—or expressing Cas12a more generally—might be open to a quick chat or could offer any suggestions to help identify what might be going wrong.

I would truly appreciate any insights or advice.
