Request for Lentiviral gRNA Vector Recommendations

Dear all,

I have generated CRISPR-inducible human iPSC lines that I plan to use for screening soon. Before proceeding, I would like to test the inducible system using a lentiviral vector expressing a specific gRNA, ideally with a selection marker such as puromycin.

If anyone has recommendations for such lentiviral gRNA vectors or is willing to share related information or resources, I would greatly appreciate your help.

Dear Yachten,

To check in ipscs you can simply use synthetic sgRNAs. You can synthesise either in vitro transcription or buy from companies such as IDT, Synthego, or Genscript. You can deliver sgRNAs using electroporation as described in the following protocol.
