U6 promoter 'G' question

Quick question: If my guide sequence starts with a 'G', do I still need to include an additional 'G' at the end of the u6 promoter? For example, if cloning into pX330, do I still need to add the additional G to the end of the CACC over hang?

I hope you are doing well. With regards to your question, if your guide sequence starts with a G, you don't have to add an additional G at the start. Best of luck!
