Seeking Recommendations for Inducible Cas9 Vectors and Nuclear Cas9 Antibodies

Dear all,

I am working on generating inducible Cas9 (iCas9) iPSC lines for a while. We have a gRNA library vector that includes mCherry and puromycin resistance markers. However, I've tested two different iCas9 vectors, and in both cases, Cas9 expression was not successfully induced.

Could anyone recommend a reliable iCas9 vector that has been effective in iPSC systems?

Additionally, I am looking for a Cas9 antibody suitable for immunofluorescence staining, specifically for detecting nuclear-localized Cas9. If you have experience with antibodies that provide clear nuclear staining, I would appreciate your suggestions.
