Many thanks to Zhang lab for developing the genome editing tools and great web resources.

I wonder if anyone could upload a protocol to produce a homemade. Stbl3 (propagated from Invitrogen C7373-03).

Dear Zhang, we used the kit you mentioned but Stbl3 cells produced with it showed strange behavior - even if you just transform linear DNA to them they will randomly recombine some regions and produce circular DNA - giving a lot of colonies on the plate. In the correct clones we also saw single nucleotide substitutions. Do you know why it might happen? Thank you!

PS. Also, we were very unhappy with Zymogen as one bottle of ZymoBroth was contaminated.
