Have a DNS server that is primary for six zones - each zone uses the
same mail server - mail1.mydomain.com. Does each zone file , in
addition to the MX record pointing to the mail server
mail1.mydomain.com, need an A record for the mail host, or is the A
record only needed in the mydomain.com zone file?

ie;

does this suffice:

MX 10 mail1.mydomain.com

or does every zone file need:

MX 10 mail1.mydomain.com

mail1.mydomain.com A xxx.xxx.xxx.xxx

or should only the mydomain.com be set up like the second example

The A record goes in the zone file for the domain the mail host is in
