When a printer prints at 300 dpi
does 300 dpi really mean dots per inch?
Or does it mean dots per square inch?
If its dpi then how much space is between
the lines and many dots print per square inch?

300 DPI means 300 dots per inch horizontally by 300 dots per inch
vertically, or 90,000 dots per square inch.
Ideally, there wouldn't be any space between the dots. In practice, it
depends on your printer. Get a magnifying glass and check out something
printed at 300 DPI.

DPI mean dots per inch. But it helps if you don't confuse dots with pixels.
A printer will need to print several dots to make on image pixel. This is
why dpi is best for referring to printer capabilities and pixels per inch
(ppi) is better for referring to image resolution and dimensions.
