Assign custom colours to groups

Hi,

I have difficulty in assigning custom colours for my groups.

The tick box is disabled. 

Do you have any advise on enabling that?

Please refer the below attached.

Also, I would like to compare multiple gene expressions within multiple groups.

If you can provide any guidance that would be greatly appreciated.
