Amalapaul's parents are Paul Varghese and Annice Paul. She has a sibling named Abijith Paul, who is an actor. Amala completed her primary education at "Nirmala Higher Secondary School," and later completed her higher secondary education at "Aluva Government Higher Secondary School."

In his Malayalam film Neelathamara (2009), it was Lal Jose who gave her maiden acting role with a supporting role, the movie was a blockbuster success. The next year, she went on to appear in several Tamil movies.

Via Prabhu Solomon's romantic comedy film called Mynaa, she became a sensation in the Tamil movie industry. She has never looked back since then and has featured in all of the South Indian language movies. However, she has primarily focused on Tamil cinema and is one of the Tamil film industry's most sought after actresses.

Deiva Thirumagal, Kadhalil Sodhappuvadhu Yeppadi, Thalaivaa, Velaiyilla Pattathari, and Amma Kanakku, among many others, are some of her finest films. Almost all the stars of the Tamil film industry have worked with her.

She appeared in the mega hit Malayalam movie Run Baby Run in the year 2012 opposite legendary Malayalam film actor Mohan Lal.

Recently she was appeared in Tamil movies, such as Thiruttu Payale 2, Ratsasan and Aadai.
