I start with registering the image stack to reduce jitter in the series ( a time series). Then, I produde a Z-Project of the stack (averaged intensity) to obtain the cleanest view of all the nerve fragments. I work with that image to produce the threshold (auto works fine) and mask.

When I played around with neural time lapse images for one of the MITx classes (Quantitative Biology Workshop, free online course and they had basically your example as a problem set), I ended up using Max intensity projection if the time series is well aligned. IIRC they used standard deviation projection to find the most frequently changing neurons. Averaging will help if you want frequently firing neurons and include some idea of intensity! However, while a neuron that fires a single time in a time series will show up just fine in a max projection, it would show up only faintly in an average, as the mean value will be lowered by all of the time the neuron is not firing.
Starting a new chat each prompt worked a few times but eventually nothing I do gets it to generate images consistently. I would expect Dall-E to be unstable but not the actual access to it VIA the ChatGPT interface. A shame too because the images generated when it does work are amazing.

Hello, I am using SNAP version 7.0.3 and with any dataset I use, if I try to create and image from a band, the program hangs on the Creating Image loading screen (regardless of using a virtual or local band). The Creating Image loading pop-up also does not close when I hit cancel. The example of this is below:

could you please explain how are you planning to use the colour data that you might extract from an image to create columns?
do you have a drawing or a sketch of the final result you are aiming for?
from your screenshot looks like you would like to use the image as a displacement map of some sort?
Its a matter of pushing the points away from the center of the circle the whiter the colors on the image, and vise versa for the black. After that all the lines can be lofted to create the end result mesh or column.
I tried your first definition, solved the extrusion problem. But it is as if the surface performs an offset at the minimum points, while in my case I want to keep the black parts of the image at a value of 0, and the parts that extrude must be those that go towards the white.

I saw this as a question originally in the Mojave forum, but the same problem applies to Catalina. If I attempt to create an image from a USB device (in my case, an SD card), Disk Utility fails to create an image and reports 'operation cancelled'.
Click on the lock at the bottom left of the screen, enter your password, then press the + button near the centre of the screen. Add Disk Utility to the list. Click on the lock again and things should work.
Here is a practical example. At the image below there is a blue zone, that is the area which I want to publish an image service for. The GSD for the whole blue zone is 50 cm. However within that area there are some other zones, in this case zone 2 and 3 where I have an additional images with resolution of 10 cm. I want a user to be able to zoom to the greatest detail possible.
So I will set the service to be cached to the actual resolution of images with 10 cm GSD. This will work fine, but the problem is that the whole blue area will be cached to GSD of 10cm even though the actual resolution for the most of the area is 50 cm. This will result in unnecessary big cache size and if the area is big enough, then the cache size can be easily 100 times greater than necessary.
I can create 2 image services. One for the high resolution imagery and other for low resolution imagery. However, when user request the tiles from the services and the map extent is located in the area of high resolution imagery, the tiles from both image services will be sent to the user from the server. The high resolution will overlap the low resolution image, but this will increase the response time, as two sets of tiles have to be downloaded.
Use 'Manage Tile Cache'. This enables you to defined not only the scales at which the imagery should be created but also Area of Interest. Run the tool twice once for the 'blue' area at the lower resolution and then again with the same cache but define AOIs for the 'red' areas. For the second time just define the higher resolution scales.
But what happens when users zooms all the way in, in the blue area where there are no high resolution tiles. Will he get the tiles which are available just digitally resampled on the fly, or no tiles will be served, so he will see white background or whatever layers is located below?
Depends on the client and the layers that have been defined. In Desktop it will resample the image and display imagery if the layer has not been defined. In web it will display blank. One alternative is to serve as an image service and turn on demand caching one. Then the server will generate the additional tiles if required.
My workaround is to firstly create the ticket. After the ticket is created I can freely attach images the way I am used to. This issue seems to have been introduced with last weeks UI update for Jira.
Experiencing the same issue. Noticed it the latter part of last week after changing to the new, however when I reverted to the older view, the issue was there as well. Does not seem to be browser related, nor was I able to find a setting that would allow/disallow pasting of sceenshots on the Create Image form.
Is there really a difference in the outcome of these two ways options? To me it feels like I am manually doing the same things that the automated wizard would do anyway. It generates snapshots, selects the kernel IDs and architectures.
Why one has a warning text and the other does not? Snapshotting a running instance is considered relatively safe, and if the AMI creation does a snapshot in the background, is it any more dangerous than doing it all by hand?
They do exactly the same if you select the no reboot option when creating the AMI directly from EC2. This basically creates a snapshot that can potentially be in a inconsistent state. For example, you are risking more having an inconsistent state if you are doing a lot of disk writes when creating the snapshot.
If you want to create a snapshot in a "consistent" state you would have to shutdown your instance first and then take a snapshot and then restart your instance. This is why the AMI creation option from EC2 is pretty useful because you don't have to stop and restart. Amazon takes care of it and also the IP address doesn't change on your instance. (If you stop/restart your instance your IP address actually changes)
I'm not really sure why Amazon doesn't have a warning if you take a snapshot directly from the volume but from the volume point of view it really doesn't matter whether the volume is being used by a running or non running instance (it only cares whether it's attach or detatch to no effect on creating snapshots)
