The Applied Biosystems Quantstudio Absolute Q is a plate-based digital PCR system that provides absolute quantification of nucleic acid target sequences. Unlike qPCR, dPCR does not rely on standard curves for quantification. It is more sensitive than qPCR and thus well-suited to tasks such as detecting rare alleles or determining copy number variance.
Training:
Before being given BioGarage access, users are required to attend a training session. Training sessions take 2-2.5 hours. To set up a training, please fill out the form below and email it to biog...@biotech.wisc.edu.
After training, instrument time can be booked using our online scheduling tool. A link to the scheduling tool can be found on the Scheduling page. BioGarage instruments are available for use on weekdays from 8 am to 4 pm. See further policies regarding instrument usage and training below.
Materials:
To run the instrument, you will need a plate kit and dPCR master mix. Individual plates can be purchased directly from the BioGarage for $150 each or you can order your own plate kits. Master mix cannot be purchased through the BioGarage and must be provided by the user. Catalog numbers for necessary materials are below.
The Applied Biosystems QuantStudio Absolute Q Digital PCR System combines the power of digital PCR (dPCR) absolute quantification with the simplicity of a qPCR workflow. With a single hands-on step that takes five minutes to complete, dPCR is now easier than ever. The QuantStudio Absolute Q system provides a high quality, consistent, and easy-to-use solution for researchers who want to join the quantification revolution.
WALTHAM, Mass., Sept. 20, 2021 /PRNewswire/ -- Thermo Fisher Scientific (NYSE: TMO), the world leader in serving science, today launched the Applied Biosystems QuantStudio Absolute Q Digital PCR System, the first fully integrated digital PCR (dPCR) system designed to provide highly accurate and consistent results within 90 minutes.
dPCR has quickly become the standard for nucleic acid quantification in oncology, cell and gene therapy development and other research applications because its absolute quantification enables higher accuracy and precision. Thermo Fisher recently acquired Combinati and its cutting-edge dPCR technology to rapidly develop and commercialize it alongside an expanding portfolio of assays.
The dPCR technology that powers the QuantStudio Absolute Q solution is already in use by biotech companies and academic institutions for longitudinal monitoring of cancer-driving mutations in liquid biopsy, precise quantification of gene inserts for cell therapy development, and copy number variation studies for earlier identification of genetic conditions in newborns.
"These early adopters of dPCR technologies have faced limitations, including error-prone operation, imprecision and poor turnaround times," said Mark Stevenson, executive vice president and chief operating officer of Thermo Fisher Scientific. "For our customers, the QuantStudio Absolute Q Digital PCR System overcomes these limitations and provides an industry-best dPCR platform to enable accelerated innovation in cancer research, rare diseases and much more."
Philip Lee, Chief Technology Officer, Senti Biosciences said "Accurate measurement of genetic biomarkers is critical to Senti Biosciences' gene circuit technology. We adopted this novel solution for its fast and simple workflow, which makes a significant difference in generating consistent and accurate data quickly. It was straightforward to implement, and new users can generate the data that we need with ease."
Unlike complex, multi-instrument workflows required for traditional dPCR, the QuantStudio Absolute Q System uses microfluidic array technology and simplified workflows, designed to improve data accuracy and consistency. Hands-on time is just five minutes, minimizing the risk of user error.

Digital PCR (dPCR) is a nucleic acid quantification technique that allows absolute quantification without the need for standard curves. Bulk reaction mix is distributed or digitized into thousands of small independent reactions so that each micro-chamber contains either one or zero copies of the target. Statistical methods are then used to calculate the original concentrations based on the number of positive and negative micro-chambers.
Eliminate cumbersome and challenging sample and reagent preparation steps from your dPCR workflow. The QuantStudio Absolute Q system requires only one hands-on step that takes under five minutes to complete with minimal technical skill.
Avoid wasting precious time moving generating micro-chambers, moving plates, and reading samples. In under 90 minutes* reagent digitization, thermal cycling, and data collection are integrated into a single system with no manual transfer steps required.
