Hello,

I hope this message finds you well.

I recently used AMBIMETER during my SAXS data analysis and obtained an ambiguity score of 0.0 for one of my samples (screenshot attached). The result also indicates that “3D reconstruction is potentially unique.”

I would like to kindly confirm whether a score of 0.0 is considered valid and acceptable, or if it might indicate a potential issue with the input data or analysis setup. I just want to ensure that the result is reliable before proceeding with further modeling.

Hi Khadija,

It is possible to have an ambimeter score of 0.0 for good data. Without seeing your data I can't say whether your particular data and analysis are valid or not, but this is a plausible result on its own.

If you're concerned about the data, make sure that you have a good Guinier fit, the MW calculated via SAXS matches the expected MW, and that your IFT/P(r) function follows best practices and well fits the data. If you haven't already you could read through the tutorials here on those subjects:

Thank you. I got a really good Guinier fit, the MW calculated via SAXS matches the expected MW, the IFT/P(r) function follows best practices, and it fits my sample well. 
I have watched the talk, but yeah, it's not a bad idea to watch it again and again.
