amyloid, presenilins and gamma secretase

just thought i'd have my two penneth.

after reading several papers arguing all angles of the
presenilins, i believe it is thus far impossible to say
with any conviction as to the true role of presenilins.
Wolfe and colleagues seem set on the idea that PS is itself
gamma secretase, although there appears to be a spatial
paradox between the intracellular locations of gamma
cleavage and PS. if pushed, i would probably go for the
suggestion that PS forms a pore in the membrane, allowing
gamma secretase access to the intermembrane cleavage site
of APP.

i look forward to any replies and alternative theories.
