Most people expressing proteins seem to use the BL21/pET system, which
relies on the T7 promoter. We have an E. coli mutant we are trying to
express Mtb genes in. As this E. coli strain does not have T7-RNA
polymerase in it, we can't use pET vectors. Nor do we want to use
pGEX vectors, since we do not want a GST fusion.
For information, the E. coli strain is MG1655 derived: MG1655
delta(lacZYA-argF)U169).
Does anybody know/have experience of his-tagged (or other non-GST
tagged) expression vectors that use other (non-T7 polymerase)
promoters?
